Wu Ningkun was born in Yangzhou, Jiangsu Province in September 1920. From 1939 to 1941, he studied in the Department of Foreign Languages of Southwest Associated University, under the guidance of Shen Congwen and Bian Zhilin, and went to the United States in 1943 to serve as an interpreter for the Air Force Division trained in China in the United States. In March 1948, after graduating from Manchester College in Indiana, Wu Ningkun enrolled in the University of Chicago for a doctorate in British and American Literature. In 1951, Lu Zhiwei, president of Yanjing University, invited Wu Ningkun to return to China for English teaching. Wu Ningkun decided to give up his doctorate and return to China as a professor.

After returning to China, Wu Ningkun successively served as a professor of British and American literature at Yanjing University, Nankai University and International Relations College. Wu Ningkun was classified as a rightist in 1957. In 1962, Wu Ningkun went to Anhui University to teach English. During the Cultural Revolution, Wu Ningkun was sent to the production team. In 1974, Wu Ningkun was transferred to Anhui Normal University to teach. After 1979, Wu Ningkun returned to the College of International Relations as a professor of English and settled in the United States after retiring in 1991.

Wu Ningkuns translated works include The Surgical Knife Is a Weapon - Biography of Bethune, The Great Gatsby, and the novels and poems of Salman Rushdie, John Steinbeck, Christopher Ethwood, Henry James, Dylan Thomas and other famous British and American writers.
